An IgG mouse monoclonal antibody (10F05) against polyethylene glycol has been generated. The antibody reacts with PEG regardless of the linker used for PEG attachment, and is able to recognize a PEGylated peptide in plasma at concentrations as low as 3 pg/mL. The antibody is readily purified in substantial quantities.

Targeting CD20 antigen on B-lymphocytes provides good opportunity for management of the target cells in patients with B-cell malignancies. By the advent of hybridoma technology, monoclonal antibodies applications exert extensive changes in medical fields such as diagnosis, treatment and purification.
The prim aim of this study was to produce monoclonal antibody against CD20 for exploitation in diagnosis.

Poly(ethylene glycol) (PEG) is often attached to compounds to increase serum half-life, reduce immunogenicity, and enhance bioavailability. Accurate and sensitive quantification of PEG conjugates is critical for product development, pharmacokinetic measurements, and efficacy studies. However, PEGylated compounds can be difficult to quantify due to epitope masking by PEG.

Vesicular stomatitis virus (VSV)-mediated cell fusion was recently proposed as an alternative fusion technique to generate monoclonal antibody (MAb)-producing hybridomas. In order to further examine this technique, we made direct comparative experiments among VSV, Sendai virus (SV) and polyethylene glycol (PEG)-mediated cell fusion in generating MAb-producing hybridomas. The distribution of immunoglobulin (Ig) isotypes secreted by the hybridomas obtained, as well as hybridoma yield and specific hybridoma yield, was compared.

Monoclonal antibodies against glutamic acid decarboxylase (anti-GAD) were modified with poly(ethylene glycol) (PEG), and the resulting conjugates were characterized. Monoclonal anti-GAD antibodies were purified from ATCC HB184 hybridoma cells by either cell culture supernatant or ascites fluid from BALB/c mice. Polyclonal rabbit IgG antibodies were also used as a model protein.
