The open reading frame 2 (ORF2) of Porcine circovirus type 2 (PCV2) encodes the major Capsid (Cap) protein, which self-assembles into virus-like particle (VLP) of similar morphology to the PCV2 virion and accumulates in the nucleus through the N-terminal arginine-rich nuclear localization signal (NLS). In this study, PCV2 Cap protein and its derivates were expressed via the baculovirus expression system, and the cellular localization of the recombinant proteins were investigated using anti-Cap mAb by imaging flow cytometry. Analysis of subcellular localization of Cap protein and its variants demonstrated that NLS mediated Cap protein nuclear export as well as nuclear import, and a phosphorylation site (S17) was identified by liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the NLS domain to regulate Cap protein nuclear export.

In clustering analysis, data attributes may have different contributions to the detection of various clusters. To solve this problem, the subspace clustering technique has been developed, which aims at grouping the data objects into clusters based on the subsets of attributes rather than the entire data space. However, the most existing subspace clustering methods are only applicable to either numerical or categorical data, but not both.

We discovered several new energetically competitive structures of NbSe2 using the multi-algorithm collaborative (MAC) crystal structure prediction algorithm combined with the density functional theory. It was found that the coordination number of Nb in NbSe2 is increased from 6 to 7, and then to 8 with increasing pressure. Furthermore, it was unexpected that an Se atom would be squeezed to the center of a cage formed by 12 other Se atoms and then have 12-fold coordination when the pressure was increased to 130.

Type 2 diabetes (T2DM) and pre-diabetes have become a major public health problem in China. We examined whether a higher dietary glycemic index (GI) or glycemic load (GL) was associated with a less favorable glucose homeostasis among Chinese adults and whether these associations were modified by their genetic predisposition or whether combined effects exist with their cereal fiber intake. Multivariable regression analyses were performed in 3918 adults aged 23-69 years for whom three 24-hour dietary recalls and information on glucose homeostasis, genetic background and potential confounders was available.

Relaxin-3 has been proposed to modulate emotional-behavioural functions such as arousal and behavioural activation, appetite regulation, stress responses, anxiety, memory, sleep and circadian rhythm. The nucleus incertus (NI), in the midline tegmentum close to the fourth ventricle, projects widely throughout the brain and is the primary site of relaxin-3 neurons. Over recent years, a number of preclinical studies have explored the function of the NI and relaxin-3 signalling, including reports of mRNA or peptide expression changes in the NI in response to behavioural or pharmacological manipulations, effects of lesions or electrical or pharmacological manipulations of the NI, effects of central microinfusions of relaxin-3 or related agonist or antagonist ligands on physiology and behaviour, and the impact of relaxin-3 gene deletion or knockdown.

A New Distance Metric for Unsupervised Learning of Categorical Data.

IEEE Trans Neural Netw Learn Syst 2016 May 9;27(5):1065-79. Epub 2015 Jun 9.
Hong Jia, Yiu-Ming Cheung, Jiming Liu
Distance metric is the basis of many learning algorithms, and its effectiveness usually has a significant influence on the learning results. In general, measuring distance for numerical data is a tractable task, but it could be a nontrivial problem for categorical data sets. This paper, therefore, presents a new distance metric for categorical data based on the characteristics of categorical values.

The fusion of monocyte/macrophage lineage cells into fully active, multinucleated, bone resorbing osteoclasts is a complex cell biological phenomenon that utilizes specialized proteins. OC-STAMP, a multi-pass transmembrane protein, has been shown to be required for pre-osteoclast fusion and for optimal bone resorption activity. A previously reported knockout mouse model had only mononuclear osteoclasts with markedly reduced resorption activity in vitro, but with paradoxically normal skeletal micro-CT parameters.
