An instrument is presented that is capable of measuring the optical spectrum (long-wave ultraviolet through short-wave mid-infrared) of fluids under a range of temperature and pressure conditions from ambient pressure up to 138 MPa (20 000 psi) and 422 K (300 °F) using ∼5 ml of fluid. Temperature, pressure, and density are measured in situ in real-time, and composition is varied by adding volatile and nonvolatile components. The stability and accuracy of the conditions are reported for pure ethane, and the effects of temperature and pressure on characteristic regions of the optical spectrum of ethane are illustrated after correction for temperature and pressure effects on the optical cell path length, as well as normalization to the measured density.

Optimizing outcome in biliary atresia (BA) requires timely diagnosis. Cholestasis is a presenting feature of BA, as well as other diagnoses (Non-BA). Identification of clinical features of neonatal cholestasis that would expedite decisions to pursue subsequent invasive testing to correctly diagnose or exclude BA would enhance outcomes.

Choline depletion is seen in cystic fibrosis (CF) and pancreatic insufficiency in spite of enzyme treatment and may result in liver, fatty acid, and muscle abnormalities. This study evaluated the efficacy and safety of an easily absorbed choline-rich structured lipid (LYM-X-SORB™ [LXS]) to improve choline status.
Children with CF and pancreatic insufficiency were randomized to LXS or placebo in a 12-month double blind trial.

Daptomycin (DAP) is a cyclic lipopeptide antibiotic used for the treatment of certain Staphylococcus aureus infections. Although rare, strains have been isolated that are DAP resistant. These strains usually have mutations in mprF, a gene encoding a membrane protein with both lysylphosphatidylglycerol (LPG) synthase and flippase activities.

Few studies have examined the association between procedural volume and clinical outcomes in heart transplantation. This retrospective study was performed on a contemporary cohort of heart transplant recipients to better elucidate the effect of transplant center volume on 1-year mortality.
Data from the Scientific Registry of Transplant Recipients were used to analyze the relationship between transplant center volume and short-term survival.

Qualification, preparation, and use of lipid compounds as analytical reference standards are daunting endeavors. The sheer vastness of the number of lipid compounds present in biological samples make it impossible to directly standardize each entity. Available lipid compounds chosen for preparation as standards are difficult to maintain as pure entities of stable concentration due to their physical and chemical interactions.

Intracellular nucleoside reverse transcriptase inhibitor triphosphate (NRTI-TP) concentrations are crucial in suppressing HIV replication. Little is known about how commonly used dual-NRTI regimens affect the intracellular levels of NRTI-TPs, the active form of these drugs. This study investigates the effect of dual-NRTI therapy in intracellular NRTI-TP levels.

A high-performance liquid chromatography (HPLC) method was developed to measure levels of d-threo-1-phenyl-2-palmitoylamino-3-morpholino-1-propanol (d-threo-PPMP) in mouse plasma and liver. d-threo-PPMP was measured by HPLC with a Luna Pheny-Hexyl column (5 microm, 250 mm x 4.6 mm) employing UV detection at 210 nm using a mobile phase of potassium phosphate buffer (20mM, pH 3.

The Aurora kinases are essential for the regulation of chromosome segregation and cytokinesis during mitosis. Aberrant expression and activity of these kinases occur in a wide range of human tumors, and lead to aneuploidy and tumorigenesis. Here we report the discovery of a highly potent and selective small-molecule inhibitor of Aurora kinases, VX-680, that blocks cell-cycle progression and induces apoptosis in a diverse range of human tumor types.
