The open reading frame 2 (ORF2) of Porcine circovirus type 2 (PCV2) encodes the major Capsid (Cap) protein, which self-assembles into virus-like particle (VLP) of similar morphology to the PCV2 virion and accumulates in the nucleus through the N-terminal arginine-rich nuclear localization signal (NLS). In this study, PCV2 Cap protein and its derivates were expressed via the baculovirus expression system, and the cellular localization of the recombinant proteins were investigated using anti-Cap mAb by imaging flow cytometry. Analysis of subcellular localization of Cap protein and its variants demonstrated that NLS mediated Cap protein nuclear export as well as nuclear import, and a phosphorylation site (S17) was identified by liquid chromatography-tandem mass spectrometry (LC-MS/MS) in the NLS domain to regulate Cap protein nuclear export.

Two strains of PRRSVs were isolated in 2006 and 2016, respectively, and designated as FZ06A and FZ16A. Through the inoculation experiments, it was shown that FZ06A caused 100% morbidity and 60% mortality, while FZ16A caused 100% morbidity without death. By using genomic sequence and phylogenetic analyses, close relation of Chinese highly pathogenic PRRSV strain with FZ06A and FZ16A strains was achieved.

Dams are universally constructed to change hydrodynamic forces and impound water in urban rivers, but few efforts have been made to expound the impact of dams on the transport of antibiotics in rivers. In this study, the main object and focus was to elucidate the intercept effect of rubber dam and the gate dam on the sediment-water phase redistribution of eight typical antibiotics along an urban river with the water replenished from wastewater treatment plants (WWTPs). The results show that the eight antibiotics were widely detected in the whole river segment, and the WWTPs were found to be the main sources.

The partitioning of ten heavy metals (As, Cd, Co, Cr, Cu, Hg, Ni, Pb, Sb, and Zn) between the water, suspended particulate matter (SPM), and sediments in seven channel sections during three hydrologic seasons in the Yangtze Estuary was comprehensively investigated. Special attention was paid to the role of tides, influential factors (concentrations of SPM and dissolved organic carbon, and particle size), and heavy metal speciation. The SPM-water and sediment-water partition coefficients (Kp) of the heavy metals exhibited similar changes along the channel sections, though the former were larger throughout the estuary.

The CuPtPd catalyst is designed and synthesized successfully via directly reducing metal ions. The composition, morphology, and structure of the as-prepared CuPtPd nanohybrids are characterized by scanning electron microscopy and transmission electron microscopy, energy-dispersive spectrometry, selected-area electron diffraction, X-ray diffraction, and inductively coupled plasma atomic emission spectrometry. By comparison of the electrocatalytic properties of the ternary CuPtPd catalyst with bimetallic catalysts, we find that ternary nanocomposites perform better electrocatalytic and antipoisoning activity toward oxidation of methanol.

Four new phenolic glycosides from Baoyuan decoction.

Four new phenolic glycosides, including two flavonoid glycosides (1 and 2) and two lignan glycosides (3 and 4), were isolated from the traditional Chinese medicine formula, Baoyuan decoction. Their structures were established by detailed analysis of the NMR and HR-ESI-MS spectroscopic data and their absolute configurations were determined by the experimental electronic circular dichroism data as well as chemical methods. Furthermore, the sources of the four new compounds were determined by the UPLC-Qtrap-MS method, which proved that 1 and 2 are originated from Glycyrrhiza uralensis, and 3 and 4 are from Cinnamomum cassia.

Clarification of the chemical composition of traditional Chinese medicine formulas (TCMFs) is a challenge due to the variety of structures and the complexity of plant matrices. Herein, an integrated strategy was developed by hyphenating ultra-performance liquid chromatography (UPLC), quadrupole time-of-flight (Q-TOF), hybrid triple quadrupole-linear ion trap mass spectrometry (Qtrap-MS), and the novel post-acquisition data processing software UNIFI to achieve automatic, rapid, accurate, and comprehensive qualitative and quantitative analysis of the chemical components in TCMFs. Photoacoustic (PA) imaging has been developed for various clinical and pre-clinical applications, and acquiring pre-beamformed channel data is necessary to reconstruct these images. However, accessing these pre-beamformed channel data requires custom hardware to enable parallel beamforming, and is available for a limited number of research ultrasound platforms. To broaden the impact of clinical PA imaging, our goal is to devise a new PA reconstruction approach that uses ultrasound post-beamformed radio frequency (RF) data rather than raw channel data, because this type of data is readily available in both clinical and research ultrasound systems.

Asymmetric cell division is a fundamental mechanism that generates cell diversity while maintaining self-renewing stem cell populations in multicellular organisms. A piece of conductive cloth has been successfully constructed from polypyrrole-coated silver nanoparticle (Ag@PPy) composites decorated on electrospun polycaprolactone (PCL) nanofibers that formed the core-shell structure of Ag@PPy/PCL@PPy via a photo-induced one-step redox reaction. The photochemical reaction method both accelerated the rate of formation of silver nanoparticles (Ag NPs) and enhanced the dispersion of Ag NPs at the surface of PCL@PPy film. The resulting Ag@PPy/PCL@PPy-based cloth was flexible enough to be cut and pasted onto a glass carbon electrode for the preparation of a biosensor.

In order to solve the bottleneck of reference standards shortage for comprehensive quality control of traditional Chinese medicines (TCMs), a series of strategies, including one single reference standard to determine multi-compounds (SSDMC), quantitative analysis by standardized reference extract (QASRE), and quantitative nuclear magnetic resonance spectroscopy (qNMR) were proposed, and Mahoniae Caulis was selected as an example to develop and validate these methods for simultaneous determination of four alkaloids, columbamine, jatrorrhizine, palmatine, and berberine. Comprehensive comparisons among these methods and with the conventional external standard method (ESM) were carried out. A simple and reliable method based on surface-enhanced Raman scattering (SERS) with a portable Raman system is described for sensitive determination of trace levels of Rhodamine B (RB) in hot sauce samples. The sodium salt of phytic acid (IP6) stabilized Au@Ag core-shell bimetallic nanoparticles are constructed and used as SERS substrate, yielding high Raman enhancement of RB. The limit of detection for RB in water is 5 nM (2 ppb), which is below China Exit and Entry Inspection and Quarantine Bureau's tolerance level of 5 ppb.

Three new flavonoid glycosides, (3R)-(+)-isomucronulatol-2'-O-β-D-glucopyranoside (1), (3R)-(-)-isomucronulatol-7-O-β-D-apiofuranosyl(1→2)-β-D-glucopyranoside (2), and (2S)-(-)-7,8-dihydroxylflavanone-4'-O-β-D-apiofuranosyl(1→2)-β-D-glucopyranoside (3), along with eight flavanones (4, 8, 10, 12, 15, 16, 21, and 24), four isoflavones (5, 11, 13, and 23), four chalcones (6, 14, 17, and 18), two isoflavans (19-20), one flavone (7), one flavonol (9), and one dihydrochalcone (22) were isolated from Baoyuan Decoction (BYD), a traditional Chinese medicine formula. The structures of the new compounds were established by detailed analysis of NMR and HRESIMS spectroscopic data, and their absolute configurations were determined by electronic circular dichroism (ECD) data. The inhibitory effects of the isolates were evaluated on nitric oxide production in lipopolysaccharide activated RAW 264.

Using discriminant analysis (DA) and principal component analysis/factor analysis (PCA/FA), we described the variations in the water quality of a constructed wetland (CW) purification system in Olympic Park supplied with reclaimed water (RW). Inositol hexaphosphate (IP6) capped silver nanoparticles (IP6@AgNPs) were fabricated as surface-enhanced Raman scattering (SERS) active substrates. SERS activity of IP6@AgNPs could be further improved via adding due amounts of Fe(3+) to form Fe(3+)-IP6@AgNPs. The mechanism of Fe(3+)-induced SERS improvement of IP6@AgNPs can be attributed to the strong interaction of IP6 and Fe(3+), which leads to controllable adjustment of the gap among neighboring nanoparticles to produce "hot spots".

WUSCHEL (WUS) is essential for preventing stem cell differentiation in Arabidopsis. Here we report that in addition to its functions in meristematic stem cell maintenance, WUS is involved in the regulation of cell division. The WUS gain-of-function mutant, stem ectopic flowers (sef), displayed elongated hypocotyls, whereas the loss-of-function wus-1 mutant had shortened hypocotyls.

Pt nanoparticles with an average size of 2-3 nm in diameter were reproducibly synthesized by reduction of H₂PtCl₆ solution containing inositol hexaphosphate (IP₆) as the stabilizing agent. Single crystals with Pt(111) faces of the resulting cubic nanoparticles were revealed by the electron diffraction pattern. The PtNPs-IP₆ nanoparticles were used to modify an electrode as a nonenzymatic sensor for H₂O₂ detection, exhibiting a fast response and high sensitivity.
